An EMS-based forward genetic mosaic screen of the X-chromosome to identify genes required for axonal maintenance in Drosophila melanogaster

Abstract
An EMS-based high-throughput forward genetic mosaic screen was performed to identify essential genes on the Drosophila X-chromosome required for axonal maintenance to uncover novel and known genes associated with neurodegenerative disorders (NDDs) and their molecular pathways. By screening for an axonal phenotype, this study found 67 stocks with a degenerative phenotype and 31 stocks with a cell lethal phenotype. With duplication mapping, 34 stocks were rescued and 4 complementation groups were formed. 3 of the 34 stocks were rescued with a small P[acman] duplication stocks, identifying a small region of interest in which the causative mutation is located. With whole genome sequencing and searching databases for human homologs this study will provide novel and known essential genes associated with NDDs. Some of these disease-associated genes might not have been associated with axonal maintenance before and may yield new disease models for genes involved in NDDs.
